For nucleic acid electrophoresis:
When high resolution is not required, either TAE or TBE buffer can be used.
For high-resolution separation of large nucleic acid fragments (>13 kb), a lower-concentration gel with TAE buffer is recommended.For small nucleic acid fragments, a higher-concentration gel with TBE buffer provides better resolution.
Note that boric acid in TBE buffer can interfere with DNA recovery and subsequent enzyme reactions; therefore, TAE buffer is recommended for DNA fragment recovery from agarose gels.
