Application For the cultivation of Saccharomyces cerevisiae and Pichia pastoris
Preparation Suspend 70.0 g in 1 L of distilled water. Heat with frequent agitation and boil to completely dissolve the powder. Autoclave at 121° C for 15 minutes. Yeast has a relatively wide tolerance to pH, so the pH of YPDA Agar is typically not adjusted. If required, the pH may be adjusted to 6.5 before sterilization by autoclaving.
Principle Peptone and yeast extract provide essential nitrogen sources, vitamins, and growth factors; D-glucose serves as the carbon source. Adenine sulfate suppresses reversion mutations at the ade1 and ade2 loci in the yeast genome, thereby reducing the proportion of red yeast colonies. This product is formulated based on a classical formulation
